What is 5-amino-1MQ?

5-amino-1MQ is a small-molecule inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). It is studied as a research tool for probing energy metabolism and fat-tissue biology (Sun et al., 2024).

What the research describes

NNMT has emerged as a target of interest in metabolic-syndrome research. A review notes that elevated NNMT is observed in liver and white adipose tissue in obesity and diabetes models, and that knocking down NNMT increased energy expenditure, reduced lipid accumulation, and improved insulin sensitivity in animal studies. NNMT consumes nicotinamide (an NAD+ precursor), so its inhibition is studied for downstream effects on NAD+ metabolism (Sun et al., 2024).
