What is ACE-031?
ACE-031 is a soluble form of the activin receptor type IIB (ActRIIB). It is designed to promote muscle growth by binding myostatin and other negative regulators of muscle mass, preventing them from signaling (Attie et al., 2012).
What the research examined
In a double-blind, placebo-controlled phase 1 study, 48 healthy postmenopausal women received a single dose of ACE-031 or placebo. ACE-031 was generally well tolerated (with injection-site erythema noted), and at the highest dose produced statistically significant increases in total lean body mass (3.3%) and thigh-muscle volume (5.1%) at day 29, along with biomarker changes suggesting effects on bone and fat metabolism (Attie et al., 2012).
The limits of the current evidence
- This was an early-phase (single-dose) study; ACE-031 is not an approved medicine, and clinical development of this class has faced safety considerations.
